主動脈是人體內(nèi)*大的動脈,把含氧的血液由心臟輸送到全身各個部位。位于血管壁的主動脈內(nèi)皮細(xì)胞(AEC)經(jīng)常暴露于高血流動力,誘導(dǎo)內(nèi)皮源性血管收縮和血管生長調(diào)節(jié)的物質(zhì)生產(chǎn)變化。AEC也調(diào)節(jié)細(xì)胞粘附分子的表達(dá)來控制和微調(diào)的炎癥反應(yīng)和纖溶系統(tǒng)。這些生理特征使得AEC廣泛被應(yīng)用于研究內(nèi)皮功能障礙機(jī)理、血管疾病和動脈硬化的發(fā)病機(jī)理、以及新的疾病的治療技術(shù)的發(fā)展。
Sciencell研究實(shí)驗室的ACE是從人主動脈分離得到的,原代凍存,冰凍運(yùn)輸。每管細(xì)胞密度超過5×10^5/ml。細(xì)胞特性經(jīng)vWF/Factor VIII和CD31 (PECAM) 免疫熒光鑒定,能夠攝取DiI標(biāo)記乙酰化低密度脂蛋白(DiI-Ac-LDL)。本細(xì)胞經(jīng)檢測不含HIV-1、HBV、HCV、支原體、細(xì)菌、酵母菌和真菌。如采用ScienCell 實(shí)驗室特制的培養(yǎng)基,可保證此細(xì)胞10倍增增殖。
